چکیده

The essence of this presentation is that under effective immunosuppression, there is a migration of sessile (passenger) leukocytes (prominently but not limited to dendritic cells) from the graft into ubiquitous recipient tissues, and replacement of these cells in the graft by similar leukocytes from the recipient. The consequence is a body wide engagement of donor and recipient cells followed, we believe, with variable donor and recipient-specific nonreactivity that mayor may not require lifetime immunosuppression for stabilityl (Fig. 1).

Cell migration and chimerism--a unifying concept in transplantation--with particular reference to HLA matching and tolerance induction.

WE recently reported that the acceptance of wholeorgan transplants connotes (and requires) a special kind of mixed chimerism involving an exchange between organ and recipient oflymphodendritic leukocytes (Fig 1 ).1 The consequence in five of five patients studied 27 to 29V2 years after kidney transplantation from HLA-mismatched donors was the diffuse presence of donor dendritic cells in the lym...

Transplant Tolerance Induction in Newborn Infants: Mechanisms, Advantages, and Potential Strategies

Although several tolerance induction protocols have been successfully implemented in adult renal transplantation, no tolerance induction approach has, as yet, been defined for solid organ transplantations in young infants. Pediatric transplant recipients have a pressing demand for the elaboration of tolerance induction regimens. Indeed, since they display a longer survival time, they are expose...

Relationship between chimerism and tolerance in a kidney transplantation model.

The persistence of donor leukocytes in recipients of organ allografts has been associated with long-term graft acceptance. However, it remains unclear whether this peripheral donor cell microchimerism plays an active role in graft acceptance or is simply a consequence of the maintenance of sufficient immunosuppression to avoid rejection. A model of kidney transplantation between swine leukocyte...

Documentation in bone-marrow-augmented organ recipients of the presence of dendritic cell progenitors of donor origin.

T HE ubiquitous presence of donor cell chimerism in the tissues of long-term successful organ allograft recipients l 3 provided quintessential support to the argument that these cells playa seminal role in allograft acceptance and in the induction of donor-specific tolerance. A finding of commensurate significance was the observation that livers (unlike other organs) when transplanted orthotopi...

Tolerance Induction Strategies in Vascularized Composite Allotransplantation: Mixed Chimerism and Novel Developments

Since the start of the clinical vascularized composite allotransplantation (VCA) era over a decade ago this field has witnessed significant developments in both basic and translational research. Transplant tolerance, defined as rejection-free acceptance of transplanted organs or tissues without long-term immunosuppression, holds the potential to revolutionize the field of VCA by removing the ne...
